目的 以超声喷涂法制备雷公藤多苷气道洗脱支架,通过星点设计-效应面法优化支架制备工艺并进行体外评价。方法 以雷公藤多苷为药物,医用硅橡胶为载体材料,制备雷公藤多苷气道洗脱支架。以雷公藤多苷的药物累积释放度(Y)为考察指标,考察硅胶用量(A)、含药量(B)、致孔剂用量(C)对药物累积释放度的影响,效应面法筛选最优处方,并对处方进行预测分析和验证,并对药物体外释放行为进行模型拟合。结果 雷公藤多苷气道洗脱支架的优选处方为:A=375 mg,B=13.60%,C=18.15%,模型预测值与实验结果值偏差为1.34%。结论 采用星点设计-效应面法得到雷公藤多苷气道洗脱支架的处方优化模型预测性良好,体外释放符合Higuchi方程,具有缓释性能。
Abstract
OBJECTIVE To prepare tripterygium glycosides air way eluting stent by ultrasonic spraying method and optimize preparation process of stent by the central composite design-response surface methodology. METHODS Tripterygium glycosides airway eluting stent was prepared by ultrasonic spraying method with medical silicone rubber as carrier material. And the effects of silicone rubber dosage (A), drug dosage (B) and pore-forming agent dosage (C) on cumulative drug release were investigated in tripterygium glycosides more cumulative drug release degree (Y) as examining index. The optimal prescription was screened by response surface method, and the predicted prescription was analyzed and verified, while drug release behavior in vitro fitted the model. RESULTS The optimal prescription of tripterygium glycosides airway eluting stent was A=375 mg, B=13.60%, C=18.15%, and the deviation between the predicted value and the experimental value was 1.34%. CONCLUSION The prescription optimization model of tripterygium glycosides airway eluting stent is obtained by the central composite design-response surface methodology. In vitro release of tripterygium glycosides airway eluting stent is consistent with the Higuchi equation and had the slow-release performance.
